Dapsone
Dapsone (diamino-diphenyl sulfone) is an antibiotic typically combined with one or two others in the treatment of leprosy and, less commonly, inflammatory conditions or other infections. Wikipedia
Some people (< 4%, depending on ethnicity) develop hypersensitivity syndrome when treated with this drug, and the syndrome itself has about a 10% mortality rate. A GWAS study of leprosy patients found two factors to be associated with increased risk for dapsone hypersensitivity syndrome: rs2844573 and the HLA-B*13:01 allele, with odds ratios of 6 and 20, respectively. On it's own, the HLA-B*13:01 allele predicts with 85% sensitivity and specificity the development of the syndrome.[PMID 24152261]
