rs760805, a SNP in the RUNX3 gene, has been reported to be the most significantly associated with risk for bladder cancer of several SNPs in this gene, based on a study of 368 Chinese patients. The adjusted odds ratio for the rs760805(A;A) genotype is 1.97 (CI: 1.44-2.69).[PMID 18684727]
Further investigation of this region in this same study indicated that 4 other SNPs in the RUNX3 gene, as well as smoking, are associated with increased risk for bladder cancer. In particular, 2 haplotypes, both containing the rs760805(A) allele, were associated with 2-3x increased risk. An individual carrying the risk forms of all 5 RUNX3 SNPs and also smoking had on average an odds ratio for bladder cancer that was 6.37 fold higher than average (CI: 4.57-8.87, p = 7.03 x 10(-28)).[PMID 18684727]
